Standardization of Sterilization Procedure for In Vitro Propagation of Capegooseberry (Physalis peruviana L)

For in vitro propagation of cape gooseberry (Physalis peruviana L.) explants were properly disinfected in order to obtain noncontaminated cultures. All the steps in standardization of micro propagation protocol was carried out in such a way as there was no chance of any contamination throughout in-vitro culture process. The explants taken from field were sterilized before their inoculation in sterile growth medium to remove dirt and dust adhering to the explants. Sodium hypochlorite (NaOCl), was tested for sterilization of nodal segments with varying times of exposure. Immersion of explants in sodium hypochlorite solution (1%) for 5minutes showed maximum survival (83.00%). Maximum mortality (46.00%) was observed when sodium hypochlorite (1%) was used for 8 minutes.
